After three days, the probe, IRDye 680 BoneTag, reveals its exact location. If you measure using intensity alone (top plate), the signal is not clear as to where it is exactly binding. In the bottom plate, we know exactly where the BoneTag is because fluorescence lifetime gives better image consistency (independent of concentration and intensity) in identifying the location of this probe (complete red coloured region with lifetime of 2ns)
